RobustPrep^TM Minicircle DNAs

Minicircles are plasmid-like, generally non-replicable supercoiled DNAs that have been showed more advantages compared with conventional plasmids, which include higher gene expression, smaller size, easier to be transfected into cells, less side effects and safer etc. Because there are no bacterial DNA sequences (e.g. Ori, antibiotic selection marker) in minicircles, they can also be called as circular expression cassettes. As a vector without viral and bacterial sequences, a vector allowing sustained high transgene expression in quiescent cells or tissues, minicircle DNA is a promising alternative to standard plasmid DNA in mammalian transgene expression studies and gene therapy.

Production of minicircles is based on site-specific recombination between attB / attP sites in the presence of integrase which is achieved in E.coli strain, followed by extraction and purification steps.

Ready-to-transfection minicircle DNAs generated from our amenable RobustPrepTM system are guaranteed free from contamination by parental plasmid DNA, miniplasmid DNA and bacterial genomic DNA.
