Conditional Gene Knockdown Plasmid Vectors _ Cre-dependent shRNA Expression RNA interference (RNAi) is a powerful strategy for gene inhibition or silence. short hairpin RNA (shRNA) is widely used in RNAi applications and it can be incorporated into the plasmid vector. shRNA consists of a sense sequence and its reverse complementary antisense sequence linked by a short loop sequence. shRNA can be constitutively and ubiquitously expressed under the control of pol II promoter (U6 or H1) or pol III promoter (e.g., CMV). However, there’s a limitation when scientists want to knock down a target gene in a time-dependent manner or in a tissue-specific manner. To overcome this limitation, a Cre-dependent genetic switch can be added to achieve time-specific or tissue-specific shRNA expression and target gene repression. For more information, please contact us at admin@rgbiotech.com. 1) Cre-Switch-ON shRNA plasmid vectors Cre-Switch-ON shRNA plasmid vectors are designed for conditional activation of shRNA expression. In the absence of Cre recombinase, a stop sequence exists upstream of shRNA expression cassette and blocks its expression. While in the presence of Cre recombinase, the stop sequence is excised and allow shRNA expression. 2) Cre-Switch-OFF shRNA plasmid vectors Cre-Switch-ON shRNA plasmid vectors are designed for conditional activation of shRNA expression. In the absence of Cre recombinase, a stop sequence exists upstream of shRNA expression cassette and blocks its expression. While in the presence of Cre recombinase, the stop sequence is excised and allow shRNA expression. |
