1. AviTag AviTag is a small peptide tag that can be fused to target proteins that allows for enzymatic biotinylation. The molecular weight of AviTag tag is 1.8 KDa. It consists of 15 amino acids [GLNDIFEAQKIEWHE]. This tag is a useful tool for biotinylation, a process of attaching biotin to proteins and other macromolecules. Biotin can be added enzymatically or chemically. Unlike chemical biotin-labeling, which is randomly occurred and may lead to protein inactivation, biotinylation using Avi-Tag technology is highly efficient and specific. In the presence of ATP and biotin, the biotin ligase (BirA) catalyzes amide linkage between the biotin and the specific lysine of the AviTag peptide. The biotinylated proteins have been applied in a wide range of areas of biochemistry and cell biology, including protein purification, detection, immobilization, visualization, tetramerization, bridging etc. |
