CEACAM5 scFv-CD28-4-1BB-CD3ζ (3rd) CAR Expression Plasmid Item No.: PCAR-100Shipping: 2 μg Storage: -20°C INTRODUCTION Chimeric Antigen Receptors (CARs) are recombinant receptors that are composed of four regions: an antigen recognition domain (ectodomain), a hinge region, a transmembrane region, and an intracellular T cell signaling domain (endodomain). CAR-T cell therapy uses T cells engineered with CARs to target specific antigen(s) for cancer immunotherapy. Carcinoembryonic antigen-related cell adhesion molecule 5 (CEACAM5), also known as Cluster of Differentiation 66e (CD66e), is a cell surface glycoprotein that belongs to a member of the carcinoembryonic antigen (CEA) family of proteins. CEACAM5 is often used as a clinical biomarker for many types of malignancies. It functions as a cell adhesion molecule and plays important roles in the inhibition of differentiation and apoptosis in colon cells. CEACAM5 protein is a target of chimeric antigen receptor (CAR) T-cells used in the treatment of relapsed/refractory CEA+ cancers. This vector contains a third (3rd) generation of CEACAM5 CAR that consists of four major components: 1) anti-CEACAM5 scFv as an extracellular antigen-binding domain, 2) a hinge region, 3) a transmembrane domain, and 4) CD28, 4-1BB (TNFRSF9, CD137) and CD3ζ (CDzeta, CD247) intracellular signaling domains.
