EGFR scFv-CD28-CD3ζ (2nd) CAR Expression Plasmid Item No.: PCAR-116Shipping: 2 μg Storage: -20°C INTRODUCTION Chimeric Antigen Receptors (CARs) are recombinant receptors that are composed of four regions: an antigen recognition domain (ectodomain), a hinge region, a transmembrane region, and an intracellular T cell signaling domain (endodomain). CAR-T cell therapy uses T cells engineered with CARs to target specific antigen(s) for cancer immunotherapy. EGFR (epidermal growth factor receptor) is a transmembrane glycoprotein that is a member of the protein kinase superfamily. It is a receptor that binds to epidermal growth factor (EGF) to induce receptor dimerization and tyrosine autophosphorylation which leads to cell proliferation. Mutations in EGFR gene are associated with lung cancer. EGFR protein is a target of chimeric antigen receptor (CAR) T-cells used in the treatment of cancers such as non-small-cell lung cancer and malignant gliomas. RGBiotech offers EGFR scFv-CD28-CD3ζ (2nd) CAR Expression Plasmid to assist CAR-T research. This vector contains a second (2nd) generation of EGFR CAR that consists of four major components: 1) anti-EGFR scFv as an extracellular antigen-binding domain, 2) a hinge region, 3) a transmembrane domain, and 4) CD28 and CD3ζ (CDzeta, CD247) intracellular signaling domains.
