GPC3 scFv-4-1BB-CD3ζ (2nd) CAR Expression Plasmid

INTRODUCTION

Chimeric Antigen Receptors (CARs) are recombinant receptors that are composed of four regions: an antigen recognition domain (ectodomain), a hinge region, a transmembrane region, and an intracellular T cell signaling domain (endodomain). CAR-T cell therapy uses T cells engineered with CARs to target specific antigen(s) for cancer immunotherapy.

GPC3 (Glypican-3) is a protein that can bind to and inhibit the dipeptidyl peptidase activity of CD26, and it can induce apoptosis in certain cell types. GPC3 with deletion mutations is associated with Simpson-Golabi-Behmel syndrome (also known as Simpson dysmorphia syndrome). GPC3 can regulate Wnt/β-catenin and Yap signaling pathways. GPC3 protein is a target of chimeric antigen receptor (CAR) T-cells used in the treatment of cancers such as hepatocellular carcinoma (HCC).
