HER2 scFv-CD28-OX40-CD3ζ (3rd) CAR Expression Plasmid Item No.: PCAR-131Shipping: 2 μg Storage: -20°C INTRODUCTION Chimeric Antigen Receptors (CARs) are recombinant receptors that are composed of four regions: an antigen recognition domain (ectodomain), a hinge region, a transmembrane region, and an intracellular T cell signaling domain (endodomain). CAR-T cell therapy uses T cells engineered with CARs to target specific antigen(s) for cancer immunotherapy. Erb-b2 receptor tyrosine kinase 2 (ERBB2), also known as HER2, is a member of the epidermal growth factor (EGF) receptor family of receptor tyrosine kinases. HER2 does not directly bind ligand. It can form homodimer or heterodimer with another ERBB member, stabilize ligand binding and enhance kinase-mediated activation of downstream signalling pathways. Overexpression of HER2 is associated with numerous cancers, including breast and ovarian tumors. HER2 protein is a target of chimeric antigen receptor (CAR) T-cells used in the treatment of cancers such as breast carcinoma and gastric adenocarcinoma. This vector contains a third (3rd) generation of HER2 CAR that consists of four major components: 1) anti-HER2 scFv as an extracellular antigen-binding domain, 2) a hinge region, 3) a transmembrane domain, and 4) CD28, OX40 (TNFRSF4, CD134) and CD3ζ (CDzeta, CD247) intracellular signaling domains.
