INTRODUCTION

Chimeric Antigen Receptors (CARs) are recombinant receptors that are composed of four regions: an antigen recognition domain (ectodomain), a hinge region, a transmembrane region, and an intracellular T cell signaling domain (endodomain). CAR-T cell therapy uses T cells engineered with CARs to target specific antigen(s) for cancer immunotherapy.

NGFR (nerve growth factor receptor), also known as p75 neurotrophin receptor (p75NTR) or CD CD271, is a member of the tumor necrosis factor receptor superfamily. NGFR is involved in pathways that promote neuronal survival and neuronal death. Diseases associated with NGFR include Prurigo Nodularis and Diabetic Neuropathy. It is a marker for cancer stem cells in melanoma and other cancers. NGFR protein is a target of chimeric antigen receptor (CAR) T-cells used in the treatment of cancers.

RGBiotech offers NGFR scFv-CD28-OX40-CD3ζ (3rd) CAR Expression Plasmid to assist CAR-T research. This vector contains a third (3rd) generation of NGFR CAR that consists of four major components: 1) anti-NGFR scFv as an extracellular antigen-binding domain, 2) a hinge region, 3) a transmembrane domain, and 4) CD28, OX40 (TNFRSF4, CD134) and CD3ζ (CDzeta, CD247) intracellular signaling domains.
