INTRODUCTION

Chimeric Antigen Receptors (CARs) are recombinant receptors that are composed of four regions: an antigen recognition domain (ectodomain), a hinge region, a transmembrane region, and an intracellular T cell signaling domain (endodomain). CAR-T cell therapy uses T cells engineered with CARs to target specific antigen(s) for cancer immunotherapy.

Receptor tyrosine kinase like orphan receptor 1 (ROR1), also known as NTRKR1, is a glycosylated type I membrane protein that belongs to the ROR subfamily of cell surface receptors. ROR1 is a receptor tyrosine kinase-like orphan receptor that modulates neurite growth in the central nervous system. ROR1 is highly expressed during early embryonic development but expressed at very low levels in adult tissues. Overexpression of ROR1 is associated with B-cell chronic lymphocytic leukaemia. ROR1 protein is a target of chimeric antigen receptor (CAR) T-cells used in the treatment of cancers such as B-cell malignancies and breast and lung cancers.

This vector contains a first (1st) generation of ROR1 CAR that consists of four major components: 1) anti-ROR1 scFv as an extracellular antigen-binding domain, 2) a hinge region, 3) a transmembrane domain, and 4) CD3ζ (CDzeta, CD247) intracellular signaling domain.
