CD22 scFv-4-1BB-CD3ζ (2nd) CAR Expression Plasmid Item No.: PCAR-021Shipping: 2 μg Storage: -20°C INTRODUCTION Chimeric Antigen Receptors (CARs) are recombinant receptors that are composed of four regions: an antigen recognition domain (ectodomain), a hinge region, a transmembrane region, and an intracellular T cell signaling domain (endodomain). CAR-T cell therapy uses T cells engineered with CARs to target specific antigen(s) for cancer immunotherapy. CD22 (also known as SIGLEC2) belongs to the SIGLEC family of lectins. It is found on the surface of mature B cells and to a lesser extent on some immature B cells. It is predicted to enable CD4 receptor binding activity. It has protein phosphatase binding activity and sialic acid binding activity. It is involved in B cell activation and functions as an inhibitory receptor for B cell receptor (BCR) signaling. It is also a regulatory molecule of endocytosis. CD22 protein is a target of chimeric antigen receptor (CAR) T-cells used in the treatment of B cell malignancies. This vector contains a second (2nd) generation of CD22 CAR that consists of four major components: 1) anti-CD22 scFv as an extracellular antigen-binding domain, 2) a hinge region, 3) a transmembrane domain, and 4) 4-1BB (TNFRSF9, CD137) and CD3ζ (CDzeta, CD247) intracellular signaling domains.
