CD24 scFv-4-1BB-CD3ζ (2nd) CAR Expression Plasmid

INTRODUCTION

Chimeric Antigen Receptors (CARs) are recombinant receptors that are composed of four regions: an antigen recognition domain (ectodomain), a hinge region, a transmembrane region, and an intracellular T cell signaling domain (endodomain). CAR-T cell therapy uses T cells engineered with CARs to target specific antigen(s) for cancer immunotherapy.

CD24 is a sialoglycoprotein expressed at the surface of most B lymphocytes and differentiating neuroblasts. It is also expressed on neutrophils and neutrophil precursors from the myelocyte stage onwards. CD24 is a cell adhesion molecule which is anchored via a glycosyl phosphatidylinositol (GPI) link to the cell surface. It contributes to a wide range of downstream signaling networks and is crucial for neural development. Cross-linking of CD24 on the surface of neutrophils induces apoptosis and this appears to be defective in sepsis. CD24 protein is a target of chimeric antigen receptor (CAR) T-cells used in the treatment of cancers.
