CD30 scFv-CD28-OX40-CD3ζ (3rd) CAR Expression Plasmid Item No.: PCAR-035Shipping: 2 μg Storage: -20°C INTRODUCTION Chimeric Antigen Receptors (CARs) are recombinant receptors that are composed of four regions: an antigen recognition domain (ectodomain), a hinge region, a transmembrane region, and an intracellular T cell signaling domain (endodomain). CAR-T cell therapy uses T cells engineered with CARs to target specific antigen(s) for cancer immunotherapy. CD30 (also known as TNFRSF8) is a member of the TNF-receptor superfamily. It is expressed by activated, but not by resting, T and B cells. TRAF2 and TRAF5 can interact with CD30, and mediate the signal transduction that leads to the activation of NF-kappaB. CD30 is a positive regulator of apoptosis, and also has been shown to limit the proliferative potential of autoreactive CD8 effector T cells and protect the body against autoimmunity. CD30 protein is a target of chimeric antigen receptor (CAR) T-cells used in the treatment of CD30+ Hodgkin lymphoma (HL) or non-Hodgkin lymphoma (NHL). This vector contains a third (3rd) generation of CD30 CAR that consists of four major components: 1) anti-CD30 scFv as an extracellular antigen-binding domain, 2) a hinge region, 3) a transmembrane domain, and 4) CD28, OX40 (TNFRSF4, CD134) and CD3ζ (CDzeta, CD247) intracellular signaling domains.
