INTRODUCTION

Chimeric Antigen Receptors (CARs) are recombinant receptors that are composed of four regions: an antigen recognition domain (ectodomain), a hinge region, a transmembrane region, and an intracellular T cell signaling domain (endodomain). CAR-T cell therapy uses T cells engineered with CARs to target specific antigen(s) for cancer immunotherapy.

CD37 is a member of the transmembrane 4 superfamily, also known as the tetraspanin family. It is a cell-surface glycoprotein that is known to complex with integrins and other transmembrane 4 superfamily proteins. It mediates signal transduction events that play a role in the regulation of cell development, activation, growth and motility, and may play a role in T-cell-B-cell interactions. CD37 expression is restricted to cells of the immune system, with highest abundance on mature B cells, and lower expression is found on T cells and myeloid cells. CD37 protein is a target of chimeric antigen receptor (CAR) T-cells used in the treatment of B cell and T cell lymphomas.
