INTRODUCTION

Chimeric Antigen Receptors (CARs) are recombinant receptors that are composed of four regions: an antigen recognition domain (ectodomain), a hinge region, a transmembrane region, and an intracellular T cell signaling domain (endodomain). CAR-T cell therapy uses T cells engineered with CARs to target specific antigen(s) for cancer immunotherapy.

CD38 (cluster of differentiation 38) is a type II transmembrane glycoprotein found on the surface of many immune cells (white blood cells), including CD4+, CD8+, B lymphocytes and natural killer cells. CD38 functions in cell adhesion, signal transduction and calcium signaling and is used as a prognostic marker for patients with chronic lymphocytic leukemia. CD38 protein is a target of chimeric antigen receptor (CAR) T-cells used in the treatment of CD38-positive hematological malignancies.

RGBiotech offers CD38 scFv-CD28-4-1BB-CD3ζ (3rd) CAR Expression Plasmid to assist CAR-T research. This vector contains a third (3rd) generation of CD38 CAR that consists of four major components: 1) anti-CD38 scFv as an extracellular antigen-binding domain, 2) a hinge region, 3) a transmembrane domain, and 4) CD28, 4-1BB (TNFRSF9, CD137) and CD3ζ (CDzeta, CD247) intracellular signaling domains.
