CD123 scFv-CD3ζ (1st) CAR Expression Plasmid Item No.: PCAR-061Shipping: 2 μg Storage: -20°C INTRODUCTION Chimeric Antigen Receptors (CARs) are recombinant receptors that are composed of four regions: an antigen recognition domain (ectodomain), a hinge region, a transmembrane region, and an intracellular T cell signaling domain (endodomain). CAR-T cell therapy uses T cells engineered with CARs to target specific antigen(s) for cancer immunotherapy. CD123 (also known as IL3RA) is an interleukin 3 specific subunit of a heterodimeric cytokine receptor. The receptor, found on pluripotent progenitor cells, induces tyrosine phosphorylation within the cell and promotes proliferation and differentiation within the hematopoietic cell lines. It can also be found on basophils, pDCs and some cDCs among peripheral blood mononuclear cells. CD123 is expressed across acute myeloid leukemia (AML) subtypes, including leukemic stem cells. It helps transmit the signal of interleukin-3, a soluble cytokine important in the immune system. CD123 protein is a target of chimeric antigen receptor (CAR) T-cells used in the treatment of acute myeloid leukemia (AML). RGBiotech offers CD123 scFv-CD3ζ (1st) CAR Expression Plasmid to assist CAR-T research. This vector contains a first (1st) generation of CD123 CAR that consists of four major components: 1) anti-CD123 scFv as an extracellular antigen-binding domain, 2) a hinge region, 3) a transmembrane domain, and 4) CD3ζ (CDzeta, CD247) intracellular signaling domain.
