CD133 scFv-CD3ζ (1st) CAR Expression Plasmid Item No.: PCAR-067Shipping: 2 μg Storage: -20°C INTRODUCTION Chimeric Antigen Receptors (CARs) are recombinant receptors that are composed of four regions: an antigen recognition domain (ectodomain), a hinge region, a transmembrane region, and an intracellular T cell signaling domain (endodomain). CAR-T cell therapy uses T cells engineered with CARs to target specific antigen(s) for cancer immunotherapy. CD133 (also known as PROM1) is a member of pentaspan transmembrane glycoproteins, which specifically localize to cellular protrusions and is often expressed on adult stem cells. CD133 is thought to function in maintaining stem cell properties by suppressing differentiation. CD133 is expressed in hematopoietic stem cells, endothelial progenitor cells, glioblastoma, neuronal and glial stem cells, various pediatric brain tumors, as well as adult kidney, mammary glands, trachea, salivary glands, uterus, placenta, digestive tract, testes, and some other cell types. CD133 protein is a target of chimeric antigen receptor (CAR) T-cells used in the treatment of advanced and metastatic malignancies. This vector contains a first (1st) generation of CD133 CAR that consists of four major components: 1) anti-CD133 scFv as an extracellular antigen-binding domain, 2) a hinge region, 3) a transmembrane domain, and 4) CD3ζ (CDzeta, CD247) intracellular signaling domain.
