BCMA scFv-CD28-OX40-CD3ζ (3rd) CAR Expression Plasmid Item No.: PCAR-089Shipping: 2 μg Storage: -20°C INTRODUCTION Chimeric Antigen Receptors (CARs) are recombinant receptors that are composed of four regions: an antigen recognition domain (ectodomain), a hinge region, a transmembrane region, and an intracellular T cell signaling domain (endodomain). CAR-T cell therapy uses T cells engineered with CARs to target specific antigen(s) for cancer immunotherapy. BCMA (also known as TNFRSF17) is a cell surface receptor of the TNF receptor superfamily. It is preferentially expressed in mature B lymphocytes, and may be important for B cell development and autoimmune response. BCMA can specifically bind to the tumor necrosis factor (ligand) superfamily, member 13b (TNFSF13B/TALL-1/BAFF), and to lead to NF-kappaB and MAPK8/JNK activation. BCMA also binds to various TRAF family members, and thus may transduce signals for cell survival and proliferation. BCMA protein is a target of chimeric antigen receptor (CAR) T-cells used in the treatment of multiple myeloma. This vector contains a third (3rd) generation of BCMA CAR that consists of four major components: 1) anti-BCMA scFv as an extracellular antigen-binding domain, 2) a hinge region, 3) a transmembrane domain, and 4) CD28, OX40 (TNFRSF4, CD134) and CD3ζ (CDzeta, CD247) intracellular signaling domains.
