DNA sequence encoding miRNA precursor can be cloned downstream of a preferred promoter or GFP reporter. After transfection into cells, miRNA precursor is expressed and subsequently processed by Dicer into mature miRNA. The functional mature miRNA is then incorporated into RNA-induced silencing complex (RISC) to interact with target mRNA molecules.
