siRNA Design & Synthesis

siRNA (small interfering RNA), also known as short interfering RNA or silencing RNA, is a kind of double-stranded RNA oligo with two overhanging nucleotides on each strand. Once enters the cell, siRNA is processed by Dicer into smaller fragments. In most cases the antisense strand is incorporated into RNA-induced Silencing Complex (RISC), followed by target mRNA binding and cleavage. siRNA is an important tool for gene loss-of-function studies.

RGBiotech’s unmatched siRNA services encompass design and synthesis of standard siRNA oligos, advanced siRNA oligos, siRNA control oligos, custom siRNA oligos. All siRNA oligos we provide are purified by HPLC.

Standard siRNA Oligos

Standard siRNA oligos are traditional unmodified double-stranded RNA molecules. For each target gene, RGBiotech design and synthesize four individual siRNAs. Customer can transfect individual siRNA into cells, or use siRNA pool to enhance interference efficiency.

Advanced siRNA Oligos

Advanced siRNA oligos are double-stranded RNA molecules with chemical modification (Please choose from Table 1). Compared with standard siRNAs, advanced siRNAs have higher stability, minimized side effects, and can exert interference in a longer period of time.

siRNA Control Oligos

siRNA controls should be included in siRNA experiments for reliable results. RGBiotech offers both negative and positive siRNA controls for RNAi experiments.

1) siRNA negative controls (Table 2)
- Universal siRNA negative control is a double-stranded RNA oligo that has no homology with mammalian genes.
- Scrambled siRNA negative control is a double-stranded RNA oligo that has the same base composition as the gene-specific siRNA but with an altered sequence.
2) siRNA positive controls (Table 3)
siRNA positive controls are published siRNAs that have effective interference ability to specific genes.

Custom siRNA Oligos

RGBiotech can also synthesize siRNA oligos (<40 bp) designed by customers. A variety of modifications (Please see Table 1) are also available to improve siRNA stability, cellular uptake, cell tracking etc.
