FLP-FRT Recombination

FLP-FRT is a site-directed recombination technology that involves the recombination of sequences between short Flippase Recognition Target (FRT) sites by the recombinase Flippase (FLP) derived from Saccharomyces cerevisiae. For FRT, the recombination site consists of two 13 bp inverted repeats separated by an 8 bp spacer [5'-GAAGTTCCTATTC-tctagaaa-GtATAGGAACTTC-3']. In the presence of FLP recombinase, cleavage occurs ahead from the asymmetric 8bp core region (5'tctagaaa3') on the top strand and behind this sequence on the bottom strand. Depending upon the orientation of the FRT sites, the intervening sequence between them can be inverted or deleted by recombination. There are several variant FRT sites, but recombination can only occur between two identical FRTs.

RGBiotech offers a series of plasmids that can be used in FLP-FRT based recombination. We also provide custom construction services to meet the different demands of our customers.
