Simian Virus 40 (SV40) large T is known to inactivate Rb through p53 leading to immortalization through cell cycle regulation. In most cases, expression of SV40 Large T antigen is sufficient to induce transformation in multiple types of primary cells. Small T Antigen could help Large T antigen to transform some cell types in certain circumstances, and in some cases expression of Small T Antigen by itself is sufficient to cause a transformed phenotype.
