Human telomerase reverse transcriptase (TERT), also known as TP2, TRT, CMM9, EST2, TCS1, hTRT, DKCA2, DKCB4, hEST2, PFBMFT1, is a catalytic subunit of the telomerase complex. Deregulation of telomerase expression in somatic cells may be involved in oncogenesis. Several studies have shown that delivery of exogenous human TERE gene into primary human cells can be used to prevent telomere shortening and overcome telomere-controlled senescence to achieve cell immortalization. Human TERT can immortalize various types of primary cells including those difficult to culture in vitro, for example, fibroblasts, retinal pigmented epithelial cells, endothelial cells, oesophageal squamous cells, mammary epithelial cells, keratinocytes, osteoblasts, and Nestin-positive cells of the pancreas. Compared with using the viral protein for cell immortalization, the advantages of using human TERT are that it does not cause cancer-associated changes or alter phenotypic properties.
