Protein kinases are groups of enzymes that are involved in protein phosphorylation, including tyrosine kinases, serine/threonine kinases, MAP2K and GSK families. Protein kinases play key roles in the signaling cascade and regulate diverse biological processes such as growth, differentiation, metabolism, apoptosis, proliferation, cell cycle and motility etc. The entire set of protein kinases encoded in the genome of an organism is termed as Kinome. The human kinome contains over five hundreds of protein kinase genes. Many of these kinases are associated with human cancer initiation and progression through ways like mis-regulated expression and/or amplification/overexpression, aberrant phosphorylation, mutation(s), chromosomal translocation, and epigenetic regulation. The development of small-molecule kinase inhibitors that can block the constitutive oncogenic activation in cancer cells has become a promising approach in cancer therapies. Protein kinases have become the second most targeted group of drug targets, after the G-protein-coupled receptors (GPCRs). So far, several small-molecule kinase inhibitors have received FDA approval for the treatment of many malignancies such as breast cancer and lung cancer, and many more are in late stages of clinical development and are likely to be approved in the coming years.
