Phage Display Plasmids Phage displasy is an in vitro screening technique that a protein of interest is fused into a phage coat protein and is displayed on the surface of the phage, which can be used for the study of protein–protein, protein–peptide, protein–DNA interactions and screening of peptides and antibodies. Phage display plasmids (phagemids) are a type of plasmids containing two distinctive features: the f1 origin of replication from an f1 phage, and the sequence encoding a truncated phage coat protein. In the absence of a helper phage, a phagemid replicates using the origin of replication (ori) for double stranded replication in its E.coli bacteria host cell. When co-transfection of a phagemid and a helper phage into an E.coli cell, the single stranded DNA replication is initiated, followed by phage particle packaging. The displaying phage particles can then be screened against targets of interest to detect interactions or screen binding candidates.
